Current Projects In My Lab Include:

  • Project #1: Clinical Implementation of Optical Genome Mapping As a Front-Line Diagnostic Test for Hematological Neoplasms
    • The Cytogenomics Lab has experienced a pivotal year marked by the successful validation and clinical launch of Optical Genome Mapping (OGM) in November 2023 for testing of acute leukemias. Moving forward, the lab plans to expand OGM testing to include myelodysplastic syndrome, relapsed acute leukemias and chronic lymphocytic leukemia, while discontinuing routine karyotype and FISH testing for these indications.
    • Overall, it is clear that OGM is revolutionizing the current landscape of genome diagnostics and has demonstrated striking clinical utility in our lab to-date, uncovering diagnostically and prognostically significant alterations previously hidden by low resolution analyses. We fully anticipate that this success will similarly translate into added clinical yield for additional indications in the coming months.
  • Project #2: Rapid targeted gene sequencing and high-resolution optical genome mapping to optimize selection of targeted chemotherapy in acute myeloid leukemia
    • The Cytogenomics Lab is also exploring Oxford Nanopore Technology (ONT) long read sequencing (LRS) for the genomic profiling of AML. ONT LRS is an alternative, relatively economical approach that can provide rapid sequencing with real-time base calling and alignment, and offers several unique advantages but most notably, the ability to reflexively enrich reads within user-defined target regions to increase the depth of coverage without preamplification or capture steps. The latter technique, known as adaptive sampling, has allowed for the rapid, high coverage profiling of commonly mutated genes in which mutations confer susceptibility to small molecule inhibitors, and to diagnostically critical genes which may confer eligibility for chemotherapy formulation or for clinical trial entry, making this a potentially faster and more versatile approach compared to short read NGS panels. Based on our results of proof of concept studies, our lab is still investigating the feasibility of implementing this assay for rapid diagnostic assessment for AML.

As a Canadian College of Medical Geneticists (CCMG) certified Cytogeneticist and Molecular Geneticist, I joined the Cytogenomics Laboratory at Vancouver General Hospital in May 2022 as the new Laboratory Director. I also support coverage in the Cancer Genetics and Genomics Laboratory at BC Cancer. I originally hail from Ontario, where I completed my MSc in Microbiology and Immunology at Queen’s University in 2009 and my PhD in 2014 from SickKids and the University of Toronto in the Department of Laboratory Medicine and Pathobiology, specializing in pediatric brain tumor research. I then went on to work as a Scientific Associate at the Department of Radiation Oncology at the Princess Margaret Cancer Centre, and then as a Senior Variant Annotation Specialist, working in the Department of Genome Diagnostics at the University Health Network. Most recently, I completed my CCMG training in Genetics and Genome Diagnostics (dual training in Cytogenetics and Molecular Genetics) at the University of Calgary. My research interests focus on test validation and variant interpretation in the context of genomic testing for somatic and inherited cancers, including approaches to employ expanded genetic testing to increase diagnostic yield and impact research potential. My recent publications have investigated methods for copy number analysis in cancer diagnostic testing, clinical implementation of circulating tumor DNA testing for lung cancer patients, clinical laboratory proficiency testing strategies for tumor molecular diagnostics, and approaches to standardization of somatic variant interpretation practices in Canadian diagnostic laboratories.

My interests lie in biomarker development, quality control, data standardization and process automation. I work on whole genome bisulfite sequencing data which allows genome wide methylation profiling. More precisely, this involves defining relevant quality control metrics, automating our quality control procedure, data management and standardizing the data format in order to allow efficient data processing and distribution. This aims to facilitate and maximize reproducibility of downstream analysis such as, for instance, biomarker discovery or sample classification.

    Research Interest

Dr. Martial Guillaud Laboratory is focused in part on developing three dimensional computer models that will help determine how populations of malignant cells progress through different disease stages and become aggressive, invasive tumours. Healthy human tissue is typically ordered and arranged in a very specific fashion, while cancerous and precancerous tissues show radical changes to this order that increase in severity as disease progresses. Dr. Guillaud’s models and simulations – and other platforms that assess the spatial arrangement of cells – are yielding new insights into how populations of cancer cells evolve and progress, helping researchers and physicians better understand cancer biology and behavior.

The lab is also focused on creating automated methods for reading clinical histopathology results. A cancer or precancer diagnosis typically depends on review of tissue cross sections by an expert pathologist. This process can be time consuming and costly. Further, for precancers, it is not always easy to accurately stage disease. Programs developed by Dr. Guillaud and his team work to objectively quantify histopathology results, rapidly measuring over 100 different features of individual cells on any tissue cross-section. The goal of these efforts is to provide more accurate and consistent diagnoses, but it also may reduce health care costs and the amount of time needed to give a diagnosis

I obtained my Bachelor of Science in Applied Chemistry and PhD in Organic Chemistry from the University of Hertfordshire (UK). I worked for the Thrombosis Research Institute and Trigen Ltd. (London, UK) as an organic chemist, designing and synthesising boron-based peptidomimetics for the treatment of thrombosis and haemophilia. I left organic chemistry to focus on analytical chemistry using liquid chromatography tandem mass spectrometry laboratory at the Thrombosis and Haemostasis Department of St. Thomas’s hospital in London, UK to develop assays for the analysis of anticoagulants in neonates. I moved to Vancouver, Canada to work for a bioanalytical services department (Cantest) developing and validating assays under Good Laboratory Practices for several years before joining Maxxam Analytics’ Forensic Equine Drug Testing department as their manager. After working for a decade in the fascinating field of forensic chemistry in the horse racing world, I decided to join the Provincial Toxicology Centre to learn more about forensic toxicology in humans.
